Immobilisation of living coral embryos and larvae

These data were collected intermittently from November 2017 to December 2019 during an exploratory investigation of the immobilisation of coral embryos and larvae. Coral gametes were collected during the 2017 and 2018 mass coral spawning events at the National SeaSimulator at AIMS (SeaSim), and were fertilised and cultured in mass larval rearing tanks. The ability of embryos and larvae to withstand being immobilised in agarose gel was tested for several species, at different developmental stages, and their ability to settle and metamorphose post immobilisation was tested. This investigation was done as a first step to determine the suitability of future microencapsulation techniques for coral larvae. Data were collected in controlled laboratory experiments in sterile petri dishes and 6-well plates in SeaSim temperature-controlled experimental rooms 2 and 9. Embryos and larvae were handled with sterile plastic pipettes and standard laboratory thermometer probes were used to track agarose gelling temperatures. Leica dissection microscopes were used to make observations of the experimental embryos and larvae, and to take photomicrographs. Survival and settlement rates of immobilised larvae were compared with control larvae using Welch’s two sample t-tests in R. Survival rates were modelled against hours post fertilisation using a logistic regression with a binomial distribution and a logit link function in R.
